In 1938 the synagogue on Gottschedstrasse in Leipzig was destroyed by the National Socialists.
There is now a memorial at the former location.
Strangers have now defaced a wall there with paint.
The perpetrators had sprayed several black and white lettering on the back of a wall, the police said on Monday.
The incident is said to have occurred sometime between Sunday afternoon and evening.
An area of around 3.5 by 1.8 meters was damaged.
The police did not provide any information on the content of the graffiti.
However, the state security has started the investigation into damage to property that is harmful to the community.
